RESUMO

Selenium (Se) is an important trace element to maintain the body's dynamic balance. Lack of Se can cause inflammation. Studies have shown that inflammation often leads to disorders of the hypothalamic-pituitary-adrenal axis, but the mechanism by which Se deficiency causes inflammation of the porcine adrenal glands is still unclear. In order to study the effect of Se deficiency on the adrenal glands of pigs, we obtained Se-deficient pig adrenal glands through a low-Se diet. The results of mass spectrometry showed that the Se content in the Se-deficient group was only one-tenth of the control group. We detected the expression of the toll-like receptor 4 (TLR4) and downstream factors by qRT-PCR and Western blotting, and found that the lack of Se affected the TLR4/NF-κB pathway. It is known that miR-155-3p, miR-30d-R_1, and miR-146b have all been verified for targeting relationship with TLR4. We confirmed by qRT-PCR that miR-30d-R_1 decreased most significantly in the Se-deficient pig model. Then we tested 25 selenoproteins and some indicators of oxidative stress. It is confirmed that Se deficiency reduces the antioxidant capacity and induces oxidative stress in pig adrenal tissue. In short, a diet lacking Se induces oxidative stress in pig adrenal tissues and leads to inflammation through the miR-30d-R_1/TLR4 pathway. This study provides a reference for the prevention of adrenal inflammation in pigs from a nutritional point of view.

Selenium (Se) is an essential trace element in organism. Se deficiency can cause many diseases, including vascular disease. Studies have shown that inflammation is the main inducement of vascular disease, microRNA (miRNA) can influence inflammation in various ways, and Se deficiency can affect miRNAs expression. To study the mechanism of aorta damage caused by Se deficiency, we constructed a Se deficiency porcine aorta model and found that Se deficiency can significantly inhibit miR-223, which downregulates the expression of nucleotide-binding oligomerization domain-like receptor family 3 (NLRP3). Subsequently, we found that in Se deficiency group, NLRP3, and its downstream (caspase-1, apoptosis-related spot-like protein [ASC], IL-18, IL-1ß) expression was significantly increased. In vitro, we cultured pig iliac endothelium cell lines, and constructed miR-223 knockdown and overexpression models. NLRP3 messenger RNA and protein levels were significant increased in the knockdown group, and decreased in the overexpression group. The results of this study show that Se deficiency in porcine arteries can induce inflammation through miR-223/NLRP3.

Hydrogen sulfide (H2S) is an air pollutant, having toxic effects on immune system. Necroptosis has been discussed as a new form of cell death and plays an important role in inflammation. To investigate the mechanism of H2S-induced immune injury, and the role of microRNAs (miRNAs) in this process, based on the results of high-throughput sequencing, we selected the most significantly changed miR-15b-5p for subsequent experiments. We further predicted and determined the targeting relationship between miR-15b-5p and TGFBR3 in HD11 through miRDB, Targetscan and dual-luciferase, and found that miR-15b-5p is highly expressed in H2S-induced necroptosis and inflammation. To understand whether miR-15b-5p/TGFBR3 axis could involve in the process of necroptosis and inflammation, we further revealed that the high expression of miR-15b-5p and the knockdown of TGFBR3 can induce necroptosis. Nec-1 treatment enhanced the survival rate of cells. Notably, H2S exposure induces oxidative stress and activates the TGF-ß pathway, which are collectively regulated by the miR-15b-5p/TGFBR3 axis. Our present study provides a new perspective for necroptosis regulated by the miR-15b-5p/TGFBR3 axis and reveals a new form of inflammation regulation in immune diseases.

Objective@#To evaluate the efficacy and safety of endovascular repair with covered stent in the treatment of Stanford B-type aortic dissection.@*Methods@#The clinical data of 40 patients with Stanford B-type aortic dissection who underwent endovascular repair with covered stent in the People's Hospital of Suqian, Nanjing Drum-Tower Hospital Group from January 2012 to December 2017 were retrospectively analyzed.The incidence of complications before and after treatment, and its influence on the serum levels of IL-1β, INF-γ, VEGF were analyzed.After one year follow-up, the changes of quality of life score before and after surgery, and the mortality, reoperation or interventional treatment within one year after operation were compared.@*Results@#Forty patients were successfully implanted with stent grafts, 5 cases of which had a small amount of endoleak, 1 case had postoperative cerebral infarction, and no complications such as paraplegia or renal failure were observed.Compared with before treatment, the serum levels of IL-1β, INF-γ and VEGF were significantly down-regulated after treatment[(3.24±0.19)ng/L vs.(2.02±1.16)ng/L, (2.65±0.23)ng/L vs.(1.14±1.27)ng/L, (1.51±0.17)ng/mL vs.(0.73±0.34)ng/mL], the differences were statistically significant(t=5.037, P=0.001; t=6.071, P=0.000; t=7.314, P=0.000). After 1 year of follow-up, the mortality rate was 5.00%(2/40), and the rate of reoperation or intervention was 5.00%(2/40). The postoperative quality of life(QLQC-30) of the patients was scored in different dimensions: emotional function, body function, role function, cognitive function and social function after 1 month, 6 months and 12 months, the differences were statistically significant(F=6.134, 7.109, 5.134, 6.925, 5.197, all P=0.000).@*Conclusion@#Endovascular repair with covered stent in the treatment of Stanford B aortic dissection can significantly improve the clinical symptoms of patients, with significant effects and less complications, and can significantly improve patients' survival and quality of life.

Objective@#In order to master the epidemic trend of corona virus disease 2019 (COVID-19) and evaluate the effect of prevention and control, we evaluate the epidemic dynamics of COVID-19 in mainland China, Hubei province, Wuhan city and other provinces outside Hubei from January 16 to February 14, 2020.@*Methods@#We collected the daily number of new confirmed COVID-19 cases by nucleic acid detection reported by the National Health Commission from January 16, 2020 to February 14, 2020. The analysis includes the epidemic curve of the new confirmed cases, multiple of the new confirmed cases for period-over-period, multiple of the new confirmed cases for fixed-base, and the period-over-period growth rate of the new confirmed cases.@*Results@#From January 16 to February 14, 2020, the cumulative number of new confirmed cases of COVID-19 in mainland China was 50 031, including 37 930 in Hubei province, 22 883 in Wuhan city and 12 101 in other provinces outside Hubei. The peak of the number of new confirmed cases in other provinces outside Hubei was from January 31 to February 4, 2020, and the peak of new confirmed cases in Wuhan city and Hubei province was from February 5 to February 9, 2020. The number of new confirmed cases in other provinces outside Hubei showed a significant decline (23% compared with the peak) from February 5 to February 9, 2020, while the number of new confirmed cases in Wuhan city (30% compared with the peak) and Hubei Province (37% compared with the peak) decreased significantly from February 10 to February 14, 2020.@*Conclusion@#The epidemic prevention and control measures taken by the state and governments at all levels have shown very significant effects, effectively curbing the spread of the COVID-19 epidemic in China.
